In this case the priority does not matter, since modem B is idle anyway and the message is sent immediately with modem B while the other messages are queued and sent with modem A.
When one of the modems is idle, Diafaan SMS Server looks for the first messages in the queue that is allowed to be routed with that modem and sends these messages. The priority only matters if there are multiple messages in the queue for the same modem or SMPP connection.

if there is 10k messages in the queue with lowest priority for modem "A", then a message come to the SQL database with highest priority and to be sent by modem "B"
let me reface the scenario,
10k messages are already queued for Modem "A" with lowest priority.
1 message come with highest priority for Modem "B"
How to handle diafaan this scenario ?

The higher priority value will be sent first, in your example that will be message B.
The priority can be any number, the messages with the highest priority number is put in front of the queue and the messages with the lowest priority are in the back of the queue. Priority 0 is the default priority in Diafaan SMS Server but you can set a custom default priority for each connector.

i just want to know to set the Priority or the message which is to be sent using MS-SQL?
for example,
Message "A" have Priority = 0
Message B"" have Priority = 1
which message will be sent first by the Diafaan Server?
0 is the highest Priority or the last number ?
